    Title: “Squeezing Droplets: Liquid-Liquid Phase Separation Under Pressure”

    Abstract: Liquid-Liquid Phase Separation (LLPS) of proteins plays critical roles in cell compartmentalization. While a growing number of proteins have been showed to spontaneously phase separate in cells, the structural and thermodynamics basis of such mesoscopic droplet-like states are yet poorly understood. I will review recent experimental observations and present a conceptual framework for rationalizing the effects of pressure and temperature on LLPS. I will also present a summary of our recent findings hnRNPA1 phase separation, which provide further insights into the self-organization principles of liquid condensates.

    “Mechanisms of CRISPR-Cas Systems”

    CRISPR-Cas systems are RNA-guided nucleases that provide adaptive immune protection against intruding genomic materials in bacteria and archaea. The complementarity of CRISPR RNA (crRNA) guides Cas protein to a target DNA and/or RNA, which then initiates strand cleavage by a specific Cas protein. Cas9, a type II CRISPR effector protein, is widely used for gene editing applications since a single guide RNA (sgRNA) can direct Cas9 to cleave DNA targets of interest.

    "Quantitative Mapping of Proteins in Live Cells Using Advanced Microscopy"

    Cells modulate their metabolism, growth and proliferation in response to external stimulii.  We use advanced fluorescence microscopy methods to characterize, in live cells, the proteins implicated in environmental regulation of cellular expression programs and cell state transitions.  The approaches we use include scanning fluctuation microscopy, fluorescence lifetime imaging/phasor and single molecule localizaiton super-resolution microscopy.

    Arsenate and phosphate are abundant on our planet and exhibit tremendous similarities: nearly identical pKa values, similarly charged oxygen atoms, and thermochemical radii that differ by only 4%. However, phosphate is indispensable whereas arsenate is toxic. Can proteins discriminate between these two anions, and how would they do so?

    "Compartment-Specific Acetyl-CoA and Acyl-Carrier Protein Metabolism in Arabidopsis"

    Plants exhibit a highly dynamic metabolome in response to different developmental and environmental cues.  Determining the organization of metabolism in time and space, especially at the subcellular level, is fundamental to understand the metabolic phenotype of a cell and precise control of carbon fluxes for engineering plants for bio-based products.  However, understanding the compartment-specific metabolism in plants has been hampered by the limitations of current analytical methods to determine the subcellular locations of metabolites.
